Paoli reflects centuries of Pennsylvania history. The community grew around an inn established in 1769 by Joshua Evans. Evans' father had purchased 500 acres from William Penn in 1719, establishing the family's land holdings near the current Paoli Post Office. Evans named his inn after General Pasquale Paoli, a Corsican general, honoring him after receiving the 45th and final toast at a Saint Patrick's Day celebration.

The inn's location on the Philadelphia and Lancaster Turnpike proved strategic, situated approximately 20 miles from Philadelphia, a day's travel for horse-drawn wagons. This location spurred the inn's success and contributed to the development of the surrounding community. The turnpike later became the Lincoln Highway and eventually US Route 30, which still runs through the area and is locally called Lancaster Pike.

Paoli holds profound Revolutionary War significance. On the evening of September 20, 1777, General Charles Grey and nearly 5,000 British soldiers launched a surprise attack on an American Patriot encampment near Paoli, commanded by General Anthony Wayne. To maintain surprise, General Grey ordered his troops to remove the flint from their muskets and use only bayonets and swords. With assistance from a Loyalist spy who provided the password, the British successfully overran American pickets. The assault resulted in approximately 201 American deaths and injuries. The Paoli Battlefield Historical Park preserves this important site.

Anthony Wayne, the American general who led the defense, was memorialized through his residence, Waynesborough, a Georgian-style mansion. The house has been converted into a museum honoring Wayne's legacy and memorializing those lost during the battle.

The arrival of the Main Line of Public Works enhanced Paoli's growth. The Philadelphia and Columbia Railroad passed through Paoli, later becoming the Pennsylvania Railroad. In the late 19th century, the Pennsylvania Railroad built suburban commuter lines, and the Main Line terminus became Paoli, giving the area its iconic identity. For generations, the Paoli Local commuter train became legendary for serving residents traveling between Philadelphia and suburban communities.

Today, Paoli maintains its connection to transportation and history. Amtrak's Keystone Service and Pennsylvanian trains stop at Paoli station, and SEPTA commuter rail provides regional transit. The Wharton Esherick Museum, showcasing the work of the renowned craftsman known as the Dean of American Craftsmen, attracts visitors exploring the area's cultural heritage.
